Tsunade, Orochimaru and Jiraiya were all characters in a folk story with being able to do magic relating to slugs, snakes and toads respectively.
In the story of the Yamata no Orochi, Susanoo kills/defeats it, like in the manga/anime where Orochimaru is sealed by Itachi's Susanoo. |

In naruto chapter 3 I think? It shows sasuke with a snake, Sakura with a slug, and naruto with a frog
